Osteoporosis is a common multifactorial disease characterized by progressively decreasing bone mineral density (BMD) and reduction bone mass associated with the increased risk of bone fractures. The most dangerous is that osteoporosis often arises and progresses in latent form for a long time and is only diagnosed after manifesting itself.
Osteoporosis affects ca. 75 million people in Europe, the USA and Japan. In 2000 some 9 million new cases of osteoporotic fractures occurred of which 1.6 million were hip fractures, 1.7 million forearm and 1.7 million clinical vertebral fractures. Europe and the Americas together accounted for 51% of all of these fractures while most of the rest occurred in the Western Pacific area and Southeast Asia.
Osteoporosis as a typical multifactorial disease is the result of the interaction between hereditary (genetic) factors and adverse environmental conditions.
It is proved that polymorphic alleles of VDR and Col1A1 genes are associated with osteoporosis development; risk of occurrence fracture and fast BMD loss at postmenopausal woman and at the patients accepting glucocorticoids.
The “OsteoGENE-M” kit is intended for fast and qualitative definition of polymorphisms of vitamin D receptor gene (VDR) and in gene encoding the basic protein of bone matrix (Col1A1), which are associated with development of osteoporosis.
Due to detection polymorphisms of these genes we can get a real opportunity to reveal the people with high risk of osteoporosis occurrence. Identifying patients at risk and taking preventive measures allows to considerably reduce the risk of bone fractures.
Analysis is intended for:
• pre - and postmenopausal woman;
• women with surgical menopause by which has been executed bilaterial ovariectomy;
• the patients accepting glucocorticoids or androgens;
• close relatives of patients with osteoporosis;
• the patients suffering malabsorption, diseases of kidneys, chronic respiratory diseases, rheumatoid arthritis and the diseases leading to partial or full immovability.
Principle of method:
Method is based on simultaneous definition of polymorphism in VDR and Col1A1 genes by means of multiplex polymerase chaine reaction (PCR) with subsequent carrying out enzymatic cleavage and polyacrylamide gel electrophoresis.
Simultaneous definition of 4 objects (SNP):
• -1997G/T and +1245G/Ò polymorphism of theCol1A1 gene
• -3731A/G and +61968Ò/Ñ polymorphism of the VDR gene
